saprophyticus (Fairbrother 1940) Shaw et al. 1951, two subspecies:
- Staphylococcus saprophyticus subsp. bovis Hajek et al. 1996
- Staphylococcus saprophyticus subsp. saprophyticus (Fairbrother 1940) Shaw et al. 1951.
Gram-positive cocci, 0.6-1.4 μm diameter, nonmotile, nonsporing.
Colonies are circular, slightly convex, S-type, pale orange pigmented, 4-9 mm in
diameter. Uniform turbidity in broth cultures followed by clearing with a fine or slightly
mucoid deposit. Facultatively anaerobic, 10-40 ºC, optimum 28-35 ºC. Grow on:
Trypticase Soy Yeast extract Agar ± 5% sheep blood, P agar, Trypticase Soy Yeast
extract Broth.
Isolated from skin & urine samples (subsp. saprophyticus) and from upper respiratory tract of healthy cows (subsp. bovis).
Present in air, soil, dust, dairy products and surface of animal carcasses. Resistant to novobiocin and lysozyme. Sensible to
lysostaphin.
Subspecies bovis - unknown (none).
Subspecies saprophyticus - urinary tract infections especially in young women. Virulence factors: adherence to uroepithelial cells,
production of a hemagglutinin, production of extracellular slime.

Positive results for benzidine test, catalase, hippurate hydrolysis, nitrate reduction, urease, acid production from: mannitol, glycerol,
N-acetylglucosamine, fructose, glucose, maltose, ribose and trehalose.
Negative results for arginine dihydrolase, clumping factor, coagulase, elastase, fibrinolysin, heat-labile and heat-stable nucleases,
hyaluronidase, hemolysin, ornithine decarboxylase, esculin hydrolysis, starch hydrolysis, utilization of citrate, H2S production, acid
production from: adonitol, dulcitol, cellobiose, raffinose, rhamnose and xylose.
Variable results for gelatinase; caseinase; beta-galactosidase, beta-glucuronidase, lecithinase, hydrolysis of Tween 20, Tween 40,
and Tween 80, alkaline phosphatase, acetoin production, acid production from: arabinose, galactose, lactose, mannose, sucrose
and turanose.
Subsp. saprophyticus
Positive results for acetoin production, benzidine test, catalase, urease, acid production from: fructose, glucose, glycerol, maltose,
sucrose, trehalose and turanose.
Negative results for coagulase, DN-ase, esculin hydrolysis, hemolysin, nitrate reduction, ornithine decarboxylase, phosphatase, acid
production from: adonitol, arabinose, arabitol, cellobiose, dulcitol, erythritol, erythrose, fucose, galactose, gentiobiose, inositol, lyxose,
mannose, melibiose, melezitose, raffinose, rhamnose, ribose, salicin, sorbitol, sorbose, tagatose and xylose.
